Background: | MRCK alpha (myotonic dystrophy kinase-related Cdc42-binding kinase alpha) is a member of the dystrophia myotonica protein kinase (DMPK) family that functions downstream of Cdc42 in actin cytoskeletal reorganization. It is a serine/threonine kinase with multiple functional domains, including phorbol ester-responsive C1 domain. Three independent coiled-coil domains and the N-terminal region preceding the kinase domain are responsible for intermolecular interactions leading to MRCK alpha multimerization. The transautophosphorylation process is critical for regulation of MRCK alpha catalytic activities. Binding of phorbol esters to MRCK releases its autoinhibition, allowing N-terminal dimerization and subsequent kinase activation. |
